It has become a nuisance with a few users that perpetually don't use the subject line. Is there a way to enforce this via policy?

(We already enforce confirm before sending, saves on knee jerk reactions.)

Nope, this is something you'd have to do on the messaging server.

Can this be done to restrict receipt of messages without a subject as well? Or can this only be done through filters on the device?

We were looking to implement this via policy to enforce this for PIN as well as other messaging services on the users devices aside from the BES.

only filters ... you'd have to create some custom layered application to check these for this functionality.
